Duvyzat vs Vyondys 53: how two US-approved medicines for Duchenne muscular dystrophy compare
A side-by-side guide for families in the Gulf, built from the US FDA labels, Gulf drug registers and official sources.
Reviewed by the Reserve Meds clinical operations team on 17 October 2026. Facts on this page come from the current US prescribing information for each medicine.
Duvyzat vs Vyondys 53: the short answer
Duvyzat and Vyondys 53 are both US FDA-approved for Duchenne muscular dystrophy, but for different patients and in different ways. No trial has compared them head to head. Your specialist decides which, if either, fits. Reserve Meds is a digital health company. We do not diagnose or prescribe.

Duvyzat and Vyondys 53 side by side
The table summarizes each current US label. Labels are long; your specialist and hospital pharmacy work from the full text.
| Feature | Duvyzat | Vyondys 53 |
|---|---|---|
| Generic name | givinostat[2] | golodirsen[3] |
| Type of medicine | histone deacetylase (HDAC) inhibitor taken by mouth[2] | antisense oligonucleotide designed for exon 53 skipping[3] |
| US-approved for | Duchenne muscular dystrophy in patients 6 years of age and older.[2] | Duchenne muscular dystrophy in patients with a confirmed mutation of the DMD gene that is amenable to exon 53 skipping. This is an accelerated approval based on an increase in dystrophin production in skeletal muscle.[3] |
| First US approval (year on the label) | 2024[2] | 2019[3] |
| How it is given | Oral suspension (8.86 mg/mL) taken twice daily with food. The dose depends on body weight.[2] | Intravenous infusion of 30 mg/kg once weekly, given over 35 to 60 minutes.[3] |
| Where and what to expect | Taken at home. Platelet counts and triglycerides are checked before starting and during treatment, and it is not started if the platelet count is below 150 x 10^9/L.[2] | Given as a weekly intravenous infusion. Kidney tests (serum cystatin C, urine dipstick and urine protein-to-creatinine ratio) are done before starting, and kidney monitoring continues during treatment.[3] |
| Boxed warning | No boxed warning. Warnings: low platelets and other blood count changes, raised triglycerides, diarrhea and other stomach effects, and QTc prolongation.[2] | No boxed warning. Warnings: hypersensitivity reactions including anaphylaxis, and possible kidney toxicity (seen in animal studies).[3] |
About Duchenne muscular dystrophy and these two medicines
Duchenne muscular dystrophy is a genetic condition that causes progressive muscle weakness and wasting and affects heart muscle. It occurs almost exclusively in males. In boys with Duchenne, muscle weakness tends to appear in early childhood and worsen rapidly.[1]
It is caused by mutations in the DMD gene, which provides instructions for making a protein called dystrophin. Muscle cells without enough dystrophin become damaged as muscles repeatedly contract and relax, and the damaged fibers weaken and die over time.[1]
The main difference
The biggest difference is who can receive them. Vyondys 53 is only for boys whose genetic test confirms a DMD mutation amenable to exon 53 skipping. Duvyzat does not depend on the type of mutation and is approved from age 6. Duvyzat is a liquid taken at home twice a day; Vyondys 53 is a weekly intravenous infusion.[2, 3]
The evidence behind each approval
Duvyzat. Study 1 (NCT02851797) was an 18-month randomized, double-blind, placebo-controlled study in 179 ambulatory boys aged 6 and older who were on a stable dose of corticosteroids. Patients on Duvyzat showed statistically significant less decline in the 4-stair climb than placebo.[2]
Vyondys 53. Study 1 (NCT02310906) measured dystrophin in muscle biopsies from 25 patients. Mean dystrophin rose from 0.10% of normal at baseline to 1.02% of normal by week 48. Continued approval may depend on a confirmatory trial showing clinical benefit.[3]
No head-to-head trial. Neither US label reports a study that compared Duvyzat directly with Vyondys 53. The trials above enrolled different patients at different stages, so their results cannot be set side by side to decide which medicine works better.
Using one after the other
Patients in both trials were on a stable dose of corticosteroids. Neither label reports a study of Duvyzat and Vyondys 53 together. Your specialist decides whether a patient's genetic result and age fit either label.[2, 3]
Safety: what the labels warn about
Both labels list warnings that the treating team monitors. The boxed warning row in the table above is the most serious level of warning on a US label.
- Duvyzat: common side effects listed on the label include diarrhea, abdominal pain, low platelets, nausea or vomiting, raised triglycerides and fever.[2]
- Vyondys 53: common side effects listed on the label include headache, fever, falls, abdominal pain, nasopharyngitis, cough, vomiting and nausea.[3]

How families in the Gulf can access Duvyzat or Vyondys 53
If the medicine is listed in your country. Ask your specialist and the hospital pharmacy about local supply first. A listing does not mean every hospital stocks it, but local supply is usually the fastest route and you may not need us.
If it is not registered. The usual legal route is a named-patient request: the treating hospital, on your specialist's written request, asks the national regulator for permission to import the medicine for one patient, using the US FDA approval as the reference. In Saudi Arabia the SFDA Special Access Program guidance covers these requests; in Bahrain, NHRA Circular 41 of 2020 sets the procedure. Other Gulf countries have their own procedures, which the hospital pharmacy confirms.[6, 7] Once approval is in place, Reserve Meds arranges supply through a US-licensed pharmaceutical wholesale partner to the hospital pharmacy.
We give you a written timeline estimate once we see the hospital file. The regulator, the treatment center and the manufacturer set their own timelines.
